Study on Apoptosis and the Expression of Senescence-Related Proteins During Aging Process in Rat Femur

Abstract
Cellular aging is accompanied by a reduction in proliferative activity and changes in gene expression. In replicative senescent cells, there is decreased expression of c-jun and c-fos and increased expression of p16 and p21. I examined the immunohistochemical expression of the c-jun, c-fos, p16INK4a and p21CPI/WAF1 in femoral articular cartilage of aging rat(8, 32 and 70 week old). I also examined the proliferating activity and apoptotic index by proliferating cell nuclear antigen and TUNEL method. The articular cartilage was morphologically divided into zone I, zone II, and zone III. The callus was also morphologically divided into osteblast, proliferating chondrocyte zone, hypertrophic chondrocyte zone and mesenchymal zone. In the articular chondrocytes, many chondrocytes of zone II were predominantly PCNA, p16, c-jun and c-fos positive(p<0.05). Whereas the expression of p21 and apoptosis were predominant in the deep portion of epiphyseal chondrocytes(p<0.05). The expression of c-jun of zone I and II was decreased during aging process significantly(p<0.05). The expression of c-fos of all zones and c-jun of zone III was decreased during aging process. The other factors showed no age-dependent expression. The decrease in the in vivo expressions of c-jun and c-fos was associated with aging process of articular cartilage. We concluded that apoptosis and the expression of senescence-related proteins during aging process in rat femur is variable with area-dependent manner.
